Laboratory test that measures the resistance of red blood cells to lysis (breakdown) when placed in solutions with decreasing concentrations of sodium chloride (hypotonic solutions). Increased fragility (lysis at weaker concentrations) indicates a defect in the cell membrane, as in hereditary spherocytosis.

Conditions & Notices
- Fasting: Not required.
- Sample type: A venous blood sample is drawn into a tube containing heparin (green tube) to prevent clotting and keep cells alive.
- Processing speed: The sample must be analyzed as soon as possible (within two hours) after collection to obtain an accurate, "fresh" result. Delay will alter the result.
- Avoid transportation: The sample should not be shaken vigorously or exposed to extreme temperatures.
