An immunohistochemical (IHC) assay is used to detect the S100 protein family, which are calcium-binding proteins. These proteins are commonly expressed in cells derived from the neural crest, including melanocytes (skin), schwann cells (nerve sheath), and glial cells (central nervous system). S100 is a sensitive but not entirely specific marker and is primarily used as an adjunct marker in the diagnosis of malignant melanoma, peripheral neuroblastomas (such as schwannoma), and certain other tumors.
